A Method for the Separation of Reticuloendothelial Cells of the Spleen and Liver

Abstract
In order to separate the reticuloendothelial cells of the liver and the spleen, the following experiment was done.
1. In normal rabbits, the minute iron particles (2-3¥ì in size) suspended in the gelatine solution was injected intravenously. And then, the livers and the spleens of these animals were excised and diced into small pieces and kept in the enzyme solution (Trisma buffer containing trypsin and hyaluronidase) of about 5 times as much as the tissue pieces in its volume. This mixture was stirred slowly at 37¨¬C for 30 minutes. This tissue suspension was slowly homogenized at 150 r. p.m. for 4 minutes, and it was filtered through 6 layers of gauze to remove clumps of the tissue of liver and spleen.
2. After filtering and passing through, the glass tube on the electromagnetic field, the reticuloendothelial cells, which had phagocyted, the injected iron particles were separated and collected purely.
